Genetic similarities among four species of the Plectranthus (L’Hér.) genus - DOI: 10.4025/actascibiolsci.v32i1.880 Biological Sciences
Bandeira, Juliana Magalhães; UFPel; Bianchi, Valmor João; UFPel; Rubin, Sílvia; UFPel; Peters, José Antonio; UFPel; Braga, Eugenia Jacira Bolacel; UFPel.
The Plectranthus genus comprises several species generally referred to as boldo, which are highly used in popular medicine due to their anti-dyspeptic, analgesic and digestion-stimulating properties. The amount of natural active principles can vary greatly in the related genotypes, which may lead to the inappropriate use of these plants. This work aimed to analyze the interspecific diversity among four species of the Plectranthus genus (P. grandis, P. barbatus, P. neochilus and P. amboinicus) and the intraspecific diversity of P. barbatus collected from different places in southern Brazil, by means of the RAPD technique. A higher genetic similarity was observed between the P. neochilus and P. amboinicus species (80%), followed by P. grandis and P. barbatus...

Genetic structure analysis of Eufriesea violacea (Hymenoptera, Apidae) populations from southern Brazilian Atlantic rainforest remnants Genet. Mol. Biol.
Sofia,Silvia H.; Paula,Francine M. de; Santos,Aline M. dos; Almeida,Fernanda S.; Sodré,Leda M.K..
Random amplified polymorphic DNA (RAPD) markers were used to analyze the genetic structure of Eufriesea violacea populations in three fragments (85.47, 832.58 and 2800 ha) of Atlantic rainforest located in the north of the Brazilian state of Paraná. A total of twelve primers produced 206 loci, of which 129 were polymorphic (95% criterion). The proportions of polymorphic loci in each population ranged from 57.28% to 59.2%, revealing very similar levels of genetic variability in the groups of bees from each fragment. Unbiased genetic distances between groups ranged from 0.0171 to 0.0284, the smallest genetic distance occurring between bees from the two larger fragments. These results suggest that the E. violacea populations from the three fragments have...

Genetic variability and vegetative compatibility of Erythricium salmonicolor isolates Scientia Agricola
Sebastianes,Fernanda Luiza de Souza; Maki,Cristina Sayuri; Andreote,Fernando Dini; Araújo,Welington Luiz; Pizzirani-Kleiner,Aline Aparecida.
The Pink Disease is caused by Erythricium salmonicolor, which attacks broad hosts, such as citrus, coffee, rubber, Eucalyptus spp. and Acacia spp., infecting mainly branches. This disease became a serious problem in Brazil, reducing the citrus production up to 10%. However the genetic diversity and compatibility of the fungus E. salmonicolor from Brazilian citrus plants is not yet evaluated. Therefore, the aims of this study were to evaluate: i) the genetic variability of E. salmonicolor in the São Paulo and Minas Gerais States by the RAPD technique, and ii) the vegetative compatibility between these isolates. After RAPD analysis, six distinct groups were observed without correlation between the isolation site or host species. In the vegetative...

Genetic variability in the endophytic fungus Guignardia citricarpa isolated from citrus plants Genet. Mol. Biol.
Glienke-Blanco,Chirlei; Aguilar-Vildoso,Carlos Ivan; Vieira,Maria Lúcia Carneiro; Barroso,Paulo Augusto Vianna; Azevedo,João Lúcio.
During some phases of of their life-cycle endophytic fungi colonize plants asymptomatically being found most frequently inside the aerial part of plant tissues. After surface disinfection of apparently healthy leaves from three varieties of mandarin orange and one tangor, and after incubation on appropriate culture medium, 407 fungal isolates were obtained, giving a total infection frequency of 81%. No fungal growth was observed from disinfected seeds, indicating that fungi are probably not transmitted via seeds. Of the fungal isolates, 27% belonged to the genus Guignardia, with 12 isolates being identified as Guignardia citricarpa Kiely, which is described as a citrus pathogen. The isolates were variable in respect to the presence of sexual structures and...

Genetic variability of native populations of Wasmannia Forel (Hymenoptera: Formicidae) and their biogeographical implications Neotropical Entomology
Souza,Ana L B; Tavares,Mara G; Serrão,José E; Delabie,Jacques H C.
Wasmannia auropunctata (Roger) and Wasmannia rochai Forel are economically important ants in the Southeast and Southwest regions of Bahia State, Brazil. Randomly Amplified Polymorphic DNA (RAPD) markers were used to distinguish and analyze the genetic variability of populations of both species from Ilhéus, Jequié, BA and French Guyana. The genetic distances among W. auropunctata and W.rochai populations (55.8-71.4%) suggest genetic differentiation among them. Wasmannia auropunctata populations from sugarcane and banana plantations in Jequié were the most distant genetically (30.1-46.3%) and may represent populations restricted to isolated fragments. The high genetic distances among W. auropunctata populations from CEPLAC experimental areas, in Ilhéus...

Genetic variability of pre and post-fragmentation cohorts of Aspidosperma polyneuron Muell. Arg. (Apocynaceae) BABT
Torezan,José Marcelo Domingues; Souza,Rogério Fernandes de; Ruas,Paulo Maurício; Ruas,Claudete de Fátima; Camargo,Elvis Hassmann; Vanzela,André Luís Laforga.
RAPD was used to access the genetic variability in Aspisdosperma polyneuron, a long-lived, late-reproducing tropical tree, and highly important for the Atlantic Forest. RAPD profiles from adults (pre-fragmentation, >300 years old) and seedlings (post-fragmentation, <<50 years old) were analyzed. Results showed a decrease of genetic polymorphism of post-fragmentation cohorts in small fragments and higher genetic diversity within population. The genetic diversity distribution suggested the establishment of fragments as protected reserves, and the transference of seedlings among fragments for conservation of A. polyneuron.

Genetic variability of Streptococcus mutans isolated from low-income families, as shown by RAPD markers BJM
Moreira,Mônica; Vicente,Vânia Aparecida; Glienke,Chirlei.
The detection of Streptococcus mutans isolates with high genetic variability in different individuals indicates the occurrence of transmissibility. In this context, nine low-income families (40 individuals in total) with similar social conditions were assessed to identify the bioserotypes of S. mutans using both biochemical and RAPD markers and to establish the degree of similarity among the intra-familial isolates. The polymorphism analysis used the coefficient of Jaccard in both a Principal Coordinates Analysis (PCO) and in a UPGMA clustering method. A total of 157 isolates were obtained from salivary samples, using the morphology of colonies recovered using MSB agar as indicators. From those, 64 were characterized biochemically as S. mutans and 10 as S....

Genetic variability of three natural populations of Maytenus aquifolium (Celesteraceae) from Telêmaco Borba, Paraná, Brazil BABT
Sahyun,Sandra Aparecida; Ruas,Eduardo Augusto; Ruas,Claudete de Fátima; Medri,Cristiano; Souza,José Roberto Pinto de; Johansson,Loana Aparecida Pereira da Silva; Miranda,Luíz Vicente; Ruas,Paulo Maurício.
Three populations of Maytenus aquifolium from Monte Alegre farm, Telemaco Borba county, Paraná, Brazil were analyzed by RAPD markers. A total of 13 primers were applied wich yielded 283 well amplified markers in all the studied populations (Mortandade, Vila Preta and Trinita), producing different values of gene diversity and polymorphic loci. The analysis of molecular variance (AMOVA) indicated that 21.77% of the genetic variation was among the population. Pairwise F ST analysis showed that the most divergent populations were closer geographically, demonstrating that other factors such as different soil types could explain this variation. Bayesian analysis for K number of clusters and the Principal Coordinate indicated that these three populations were...

Genetic variation among pathogens causing "Helminthosporium" diseases of rice, maize and wheat Trop. Plant Pathol.
WEIKERT-OLIVEIRA,RITA C. B.; RESENDE,M. APARECIDA DE; VALÉRIO,HENRIQUE M.; CALIGIORNE,RACHEL B.; PAIVA,EDILSON.
Twenty isolates of four fungal species, agents of "Helminthosporium" diseases in cereals, were collected from different regions: nine Bipolarisoryzae isolated from rice (Oryza sativa), seven B.sorokiniana from wheat (Triticum aestivum), two B. maydis, and two Exserohilumturcicum from maize (Zea mays). The strains were compared by PCR-RFLP and RAPD analysis. Size polymorphism among the isolates in the ITS region comprising the 5.8 S rDNA indicated genetic differences among the isolates, while a UPGMA phenogram constructed after the digestion of this region with restriction enzymes showed inter- and intra-specific polymorphism. The RAPD profiles indicated an expressive level of polymorphism among different species, compared with a low level of polymorphism...

Genetic variation among species, races, forms and inbred lines of lac insects belonging to the genus Kerria (Homoptera, Tachardiidae) Genet. Mol. Biol.
Ranjan,Sanjeev Kumar; Mallick,Chandana Basu; Saha,Dipnarayan; Vidyarthi,Ambarish S; Ramani,Ranganathan.
The lac insects (Homoptera: Tachardiidae), belonging to the genus Kerria, are commercially exploited for the production of lac. Kerria lacca is the most commonly used species in India. RAPD markers were used for assessing genetic variation in forty-eight lines of Kerria, especially among geographic races, infrasubspecific forms, cultivated lines, inbred lines, etc., of K. lacca. In the 48 lines studied, the 26 RAPD primers generated 173 loci, showing 97.7% polymorphism. By using neighbor-joining, the dendrogram generated from the similarity matrix resolved the lines into basically two clusters and outgroups. The major cluster, comprising 32 lines, included mainly cultivated lines of the rangeeni form, geographic races and inbred lines of K. lacca. The...

Macrophomina phaseolina has been considered one of the most prevalent soybean (Glycine max) pathogens in Brazil. No genetic resistance has been determined in soybean and very little is known about the genetic diversity of this pathogen in tropical and sub-tropical regions. Fifty-five isolates from soybean roots were collected in different regions and analyzed through RAPD for genetic diversity. The UPGMA cluster analysis for 74 loci scored permitted identification of three divergent groups with an average similarity of 99%, 92% and 88%, respectively. The three groups corresponded to 5.45%, 59.95% and 34.6%, respectively of all isolates used. A single plant had three different haplotypes, while 10.9% of the analyzed plants had two different haplotypes. In...

Geographic pattern of genetic diversity in natural populations of Rosewood (Aniba rosaeodora), in the Central Amazonia Acta Amazonica
Santos,Ronaldo Pereira; Ângelo,Paula Cristina da Silva; Sampaio,Paulo de Tarso Barbosa; Quisen,Regina Caetano; Leite,Ângela Maria Conte; Oliveira,Cristiane Lopes de.
Rosewood (Aniba rosaeodora Ducke, Lauraceae) is an Amazonian evergreen tree and a source of the purest linalool, the main component of its essential oil, which is very valuable in the international perfumery market. After decades of over-exploitation it is currently considered as threatened. We evaluated the genetic diversity and its distribution in four populations in Central Amazonia. Thirty-five reliable RAPD markers were generated, of which 32 were polymorphic (91.4%). Variation was higher within the populations (76.5%; p < 0.0001) and geographic distribution contributed to population differentiation (23.4%; p < 0.0001). The Amazon River had a small influence on gene flow (3.3%; p < 0.0001), but we identified evidence of gene flow across the...

Stringari,Danyelle; Glienke,Chirlei; Christo,Daniel de; Maccheroni Jr.,Walter; Azevedo,João Lucio de.
RAPD markers were used to investigate the distribution of genetic variability among a group of Guignardia citricarpa, G. mangiferae, and Phyllosticta spinarum isolates obtained from several hosts in Brazil, Argentina, Mexico, Costa Rica, Thailand, Japan, United States and South Africa. Pathogenic isolates G. citricarpa Kiely (anamorph form P. citricarpa McAlp Van Der Aa) are the etiological agent of the Citrus Black Spot (CBS), a disease that affects several citric plants and causes substantial injuries to the appearance of their fruits, thus preventing their export. Several previous studies have demonstrated the existence of an endophytic species with high morphological similarity to the causal agent of CBS that could remain latent in the same hosts....
